Reasons: the sublimation stage heats up too fast or the sublimation stage has not yet ended before entering the analytical stage, resulting in high temperature, liquid evaporation into gas, resulting in volume reduction; Or dry products dissolve into the liquid, resulting in volume reduction, serious melting will cause bubbling phenomenon.
Solution: Reduce the temperature of sublimation stage, extend the time of sublimation stage and increase the vacuum degree of freeze-dried box, and control the product temperature below the eutectic point temperature of more than 5℃.
Cause: Incomplete drying and remaining ice crystals.
Solution: By extending the sublimation drying time or increasing the sublimation temperature to solve.
The product has no fixed shape after freeze-drying, and even the dried product is taken to the outside of the container by sublimation air
Reason: The concentration of solid matter in the product is too low to form a skeleton.
Solution: Increase the solid concentration.
Reason: The general solid substance concentration between 2% and 30% is appropriate, and the best concentration is 10% to 15%. The concentration of liquid medicine is too high, and the crystal structure formed during freezing is too dense, resulting in small pores and poor ventilation.
Solution: Reduce the concentration of the liquid or reduce the sublimation stage temperature appropriately.
Reason: The surface of the product forms an airtight glass-like structure during pre-freezing, but the heat treatment is not done. After sublimation, the product heats up, some products melt and shrink, and the contraction of the product causes the surface layer to split, so the sublimation of the lower layer can proceed normally.
Solution: Do heat treatment when pre-freezing.
